During maternal sepsis, maternal inflammation causes brain damage in the offspring, but the mechanisms involved are unknown. We have previously demonstrated that during maternal sepsis, as in the adult model of systemic lipopolysaccharide (LPS) administration, maternal brain inflammation is accompanied by hippocampal neuronal apoptosis. In the present study, in order to examine the mechanisms involved in the brain damage associated with maternal sepsis, the role of cytokines in the damage induced by maternal sepsis was examined in the neonatal mouse model. Maternal LPS injection caused a significant increase in the concentration of cytokines in the maternal plasma and cerebral cortex and a similar increase in the amount of apoptotic cells in the neonatal brain. Moreover, maternal LPS injection caused a significant increase in the levels of caspase-3 in the neonatal brain. This increase in caspase-3 in the neonatal brain was accompanied by a significant increase in the concentrations of the pro-inflammatory cytokines IL-1beta and IL-6 in the maternal plasma, which persisted for at least 24 h. Interestingly, maternal LPS administration significantly increased the production of chemokines in the maternal plasma and the neonatal brain, which was abolished in the presence of a blocking monoclonal antibody against TLR-4 or a broad spectrum inhibitor of G-protein-coupled receptors.
